The world now has a new variant coronavirus mutant spreading from one continent to another — and it looks like it is a tough one to stop.

The World Health Organization declared recently the variant that emerged in India, known as B1617, is officially a “variant of concern” because of its increased transmissibility. Last week, NPR published a story explaining why the variant B1617 could be the fastest-spreading mutant on the planet.

Like other variants of concern, B1617 has more than a dozen mutations. But two mutations in particular have been most disturbing and of great concern, said Ravindra Gupta, a microbiologist at the University of Cambridge. These mutations are found on an important part of the virus — where the immune system attacks.

Each of these mutations — or similar versions of them — have cropped up separately in other parts of the world, including one in California and one in Brazil. Previous studies have shown that these mutations, individually, give the virus an advantage against our immune system. It reduces the ability of our antibodies to fight off the virus.

At the same time, researchers at Emory University performed similar studies with antibodies taken from 15 people who have been vaccinated with either the Pfizer or Moderna shots. These studies show that the antibodies could still neutralize the B1617 variant, but the potency of the antibodies dropped by about sevenfold on average, the authors reported.

Over 273 million doses of COVID-19 vaccines were administered in the United States from December 14, 2020, through May 17, 2021. During this time, VAERS ( Vaccine Adverse Event Reporting System) received 4,647 reports of death among people who received a COVID-19 vaccine. CDC and FDA physicians review each case report of death as soon as they are notified and CDC requests medical records to further assess the reports of deaths.

There were also reported cases where patients experienced hypersensitivity or allergic reactions, related to neurology, cardiology, haematology (blood disorders) and dermatology.

Certain viruses, especially coronaviruses including SARS-CoV (that causes SARS), MERS-CoV (causes MERS), and SARS-CoV-2 (which causes COVID19), require TMPRSS2 for their entry into the body. Recent studies have shown that SARS-CoV-2 (in addition to SARS-CoV and MERS-CoV) require ACE2 (the main receptor) as well as TMPRSS2 for entry into epithelial cells.

Both ACE2 and TMPRSS2 are expressed in our nasal, bronchial, and gastrointestinal epithelium. TMPRSS2 activates; or primes, the spike protein domain (a key glycoprotein found on coronaviruses) which leads to the virus fusing to the respiratory epithelia on the cell surface through binding to ACE2. As TMPRSS2 is a serine protease, it primes the spike-domain (S) of SARS-CoV-2 by cleaving as the S1/S2 sites.

In this study, Caviarlieri supplements were given to 5 clinical subjects who are “TMPRSS2-high expressors”. After 30 days of taking Caviarlieri supplements, their TMPRSS2 receptors were quantitated. All of them were found to have significantly reduced expression of the TMPRSS2 receptor that is known for activating the spike of the virus.

One of clinical subjects, who is an orthopaedic physician has shown a significant reduction of the TMPRSS2 levels by 75-fold with Caviarlieri intake and he was Covid-19 Negative even though all members of his family and his collaborators were found positive for Covid-19. Even after repeated molecular tests for Covid-19, this clinical subject was tested and found to be Covid-19 Negative.

This study suggests that Caviarlieri can potentially act protectively against Covid-19 infection by modulating the expression of TMPRSS2 receptor. Further research studies are being conducted to validate the above findings.
